So after a day of almost continuous Gram Staneing I feal that I am (finaly) semi competent at performing this test, Hooray! I am however frustrated by streak plastering ability! Whenever I perform this test The result is generally a few isolated colonies predominated by large lines of indefinable colonies that cannot be isolated. Is the result as you can imagine does not look good and is a pain to deal with. Is there anything that I can do to deal with this frustrating issue?

I can demonstrate again for you next week, if you’d like? Do bear in mind that high motility will always make single colony isolation nearly impossible, so it might have very little to do with your technique, especially considering the types of organisms you’re isolating.

streak more lines also possibly? 4/5 on each side of the plate and then into the centre 🙂
If you’re just getting large lines of colonies grouped too closely it may be worth flaming (and cooling!) your loop between each streak – you’re aiming to create a dilution and by flaming in between you create more of a dilution….
My problem is that post flaming (as you described) I still seem to manage to get large lines of colonies. Hopefully i can figure out what I am doing wrong after Nicola has govern a demonstration
